As for the sonic hedgehog signal pathway, smoothened (SMO) receptor of is activated when Sonic hedgehog is united with Patched1 (PTCH1), the receptor in the cell membrane. It is known that the activated SMO inhibits SUFU, the negative regulator of hedgehog signaling leading to proliferation / migration / invasion of cancer cells by GLI2 activity, the transcription factor.

The HH201 GLI1 inhibitor effectively blocks the transcriptional activity of GLI1, the major activating factor of the hedgehog signaling pathway, inhibiting the expression of the target gene induced by the hedgehog. This allows useful application as a treatment for general cancer, resistant cancer, recurrent cancer, or metastatic cancer.
